PT-141 is the research name for bremelanotide, a medication that works on the brain to increase sexual desire. An FDA-approved version (Vyleesi) treats low sexual desire in women before menopause, and providers also prescribe it off-label for men and for women after menopause. MultiGen Wellness prescribes compounded PT-141 as an injection, as a nasal spray with oxytocin, or as a dissolving tablet combined with tadalafil and oxytocin.

How PT-141 differs from Viagra and Cialis

Sildenafil (Viagra) and tadalafil (Cialis) increase blood flow. They help the body respond, but they don’t create desire, so they work best when interest is already there. PT-141 works on the other side of the equation: it acts on melanocortin receptors in the brain that are involved in sexual motivation. That’s why some products combine the two, so one ingredient supports desire and the other supports blood flow.

What the studies show

Bremelanotide was approved based on two Phase 3 trials called RECONNECT, with about 1,200 premenopausal women. Compared with placebo, it modestly improved desire and reduced distress about low desire. It did not significantly increase the number of satisfying sexual encounters, so it helps some people more than others.

Earlier, smaller studies in men with erectile dysfunction found better erectile response, including in some men who hadn’t done well on sildenafil. Those studies were not large enough for FDA approval in men.

Side effects

The Vyleesi label limits use to one dose in 24 hours and no more than 8 doses a month.

Who should not use PT-141

Low desire has causes worth checking

Low libido is often hormonal. In men it’s one of the most common signs of low testosterone. In women, falling estrogen, progesterone and testosterone around menopause play a big part. Thyroid problems, high prolactin and some antidepressants can also lower desire. Labs can show whether a hormone problem is behind it, which may matter more than any single medication. See testosterone therapy for men, women’s hormone therapy and our sexual health page.

Frequently asked questions

Does PT-141 work for men?

It is not FDA-approved for men, but smaller studies in men with erectile dysfunction found it improved erections, and providers prescribe it off-label. It is often combined with tadalafil for men.

How long does PT-141 last?

Effects usually start within about an hour and can last several hours. The label calls for no more than one dose in 24 hours.

Can I take PT-141 with Cialis?

Yes, they are often combined, and one of the forms we prescribe is a single tablet with both. Your provider checks your blood pressure and heart history first, since both can affect blood pressure.

Is PT-141 the same as Melanotan?

They are related. PT-141 was developed from melanotan II research, but it was designed to focus on desire, not tanning. Melanotan II is not an approved drug.
